Hello,I am Hyung-Taek Lim working for pharmaceuticals in Korea.We have glimepiride /metformin HCl immediate and slow-release combination tablets.I have found several academic analytical papers regarding Simultaneous determination of metformin and glimepride in pharmaceutical dosage by HPLC but there is no clear about dissolution medium for simultaneous release of glimepiride /metformin HCl combination tablets.if someone who had or have been working with this Please let me know.Thanks for your help.Sincerely. A quick check of the USP does not show a monograph for a combination product such as this, but there is valuable information there: dissolution of metformin HCl extended release tablets is done in pH 6.8 buffer (App 2 at 100 rpm or App 1 at 100 rpm, depending on potency). Dissolution for glimepiride tablets uses pH 7.8 buffer with App 2 at 75 rpm.If the metformin HCl conditions are appropriate for your product, then I would check solubility of glimepiride at pH 6.8, and try performing the dissolution under the conditions specified for the metformin product.
